丹参对兔视网膜光损伤保护作用的实验观察 被引量:4

摘要 目的观察丹参对兔眼光损伤视网膜的保护作用方法健康青紫兰兔8只,随机分为对照组、实验组,每组4只。在循环光环境适应7d,光照前暗适应24h:散大瞳孔至8mm后,14500lx光照30min,然后暗适应30min,在光照后1h,对照组静脉推注0.85%生理盐水20ml;实验组静脉推注丹参注射液20ml(冻干粉1.0g/kg+0.85%生理盐水)。光照后48h取视网膜制成超薄切片,电镜观察视网膜形态结构变化。结果实验组视网膜组织结构清楚,细胞形态规整,对照组光感受器外节膜盘崩解非常明显,外核层细胞排列紊乱,核固缩.色素上皮层细胞核染色质边集,呈现细胞凋亡的早期改变,线粒体空泡变性。结论丹参对兔眼光损伤视网膜具有保护作用。 Objective To observe the protective effect of salvia mihiorrhiza on retinal in rabbits against photic injury. Methods 8 healthy gray rabbits were divided into 2 groups randomly: the control group and the experiment group, with 4 rabbits in each group. The rabbits in each group were subjected to intermittent light exposure in a daily schedule of 12 hours of light alternating with 12 hours of darkness for 7 days followed hy a 24- hour darkness. Before the experiment, mydriatics was used till the pupil to 8 mm in diameter. The rabbits were given light exposure of 30 minutes ( 14 500 lx) , then 30 minutes of darkness. An hour later, the eyes were administered with salvia mihiorrhiza 1.0 g/kg + 0.85% isotonic NaCl 20 ml, 0.85% isotonic NaCl 20 ml intravenously in the experiment and control group, respectively. After 48 hours of light exposure, the eyes were removed to make retinal uhrathin sections for observation under electron microscopy. Results Retinal structures had clear layers and regular cell shape. However, the rabbits in the control group, the outer segments of photoreceptor were arranged in an irregular order, and the stack structure of membranous disks were disorganized, and cells of the outer nuclear layer were in a disorganized pattern with karyopyenosis. The nuclear chromatin margination emerged in purpurogenous membrane, presenting early changes of apoptosis, and the mitochondria was vesiculated. Conclusion Salvia mihiorrhiza protects retinal in rabbits against photic injury.
