复合酶法提取海带多糖的研究 被引量:34

摘要 采用复合酶法(纤维素酶、果胶酶、木瓜蛋白酶,比例为2:1:1)从海带中提取3种多糖,并分析了不同加酶方式对海带多糖提取率和含量的影响。结果表明:分步加酶法(先加纤维素酶:pH值为5.0,温度55℃,浸提10min;然后加果胶酶:pH值为4.2,温度50℃,浸提2h;最后再加木瓜蛋白酶:pH值为6.0,温度55℃,浸提2h),比同步加酶法(pH值为5.5,温度55℃,浸提4h 10min)3种海带多糖和总多糖的提取率及多糖含量都有显著地提高。分步加酶法提取的褐藻糖胶、褐藻淀粉、褐藻胶及总多糖的提取率分别为3.49%、2.05%、6.00%和11.54%,比同步加酶法分别提高了45.42%、37.58%、22.95%和31.58%;分步加酶法提取的褐藻糖胶、褐藻淀粉及褐藻胶的含量分别为87.1%、59.3%和88.6%,比同步加酶法分别提高了11.95%、4.77%和5.23%。 Compound enzymes of cellulose, pectolase and papain with ratio of 2 : 1 : 1 were used to isolate polysaccharides from laminaria, and influence of two different ways of addition of compound enzymes on polysaccharide extracting rate and polysaccharide content were studied. The results showed that the way of step by step addition of compound enzymes (first cellulose, then pectolase, and finally papain) could achieve higher extracting rate and polysaccharide content than that of one step. The extracting rates of fucoidan, laminarian starch, algin and total polysacchoride through the way of step by step were 3.49%, 2.05%,6.00% and 11.54%, increased by 45.42%,37.58%,22.95% and 31.58% respectively, compared with that of one step. Meanwhile,the results also demonstrated that polysaccharide contents of fucoidan,laminarian starch, and algin through step by step were 87.1%.59.3% and 88.6%, increased by 11.95%,4.77% and 5.23%,respectively.
