低强度脉冲超声波促进皮肤成纤维细胞增殖的实验研究 被引量:8

摘要 目的:研究不同辐照参数(频率、声强、辐照时间)低强度脉冲超声波(LIPUS)对成纤维细胞增殖的影响。方法:采用MTT法测定频率1MHz、1.2MHz、1.7MHz,每一频率下四种声强的LIPUS,同一声强分别辐照5min、10min、15min、20min和30min体外培养的人真皮成纤维细胞24h后的生长增殖情况,筛选出促进成纤维细胞生长的最适宜频率、声强和辐照时间。结果:①在MTT法测定细胞活性中三种频率(1MHz、1.2MHz、1.7MHz),每一频率下四种声强的LIPUS辐照成纤维细胞5min,24h后吸光光度值均增加,辐照成纤维细胞10min,24h后吸光光度值增加最高,较对照组都有显著性差异(P<0.01)。超过一定辐照时间后,吸光光度值下降,且随着辐照时间的延长,吸光光度值下降显著。②频率1MHz、声强0.1W/cm2、辐照时间10min下的细胞相对成活率最高,与对照组比较差异有统计学意义(P<0.01)。结论:不同频率、不同声强的LIPUS辐照一定时间均能促进人真皮成纤维细胞增殖,超过一定辐照时间则会抑制细胞生长。而频率1MHz、声强0.1W/cm2、辐照时间10min为促进成纤维细胞增殖较为适宜的辐照参数。 Objective:To observe the effects of different low intensity pulsed ultrasound (LIPUS) on proliferation of fibroblast and to screen out the optimum parameters of LIPUS. Methods:LIPUS of different frequency(1,1.2,1.7MHz)and different intensity was used to stimulate the cultured human fibroblast cells for 5 min, 10 min,20 min and 30min,respectively,and examination of cell proliferation by MTr after 24h.Results:①Optical absorption value of ceil activity in MTT assay increased significantly in the ceil culture with LIPUS of different frequency (1,1.2,1.7MHz) and four intensity of each frequency treatment for 5,10 minutes,as compared with the control group (P〈0.01). With a peak detected in the cell culture with LPUS treatment for 10 minutes. But optical absorption value decreased with longer treat time, optical absorption value decreased significantly in human skin fibroblasts with LIPUS stimulation for more than 30 minutes (P〈0.01),as compared with the control group.②The peak relative survival rate detected in the ceil culture with LIPUS of 1MHz,0.1W/cm^2 treatment for 10 minutes. Conclusion: Stimulation with LIPUS could induce cell proliferation at a shorter time and could inhibit cell growth in vitro at a longer time. 1MHz,0.1W/cm^2 treatment for 10 minutes is the better parameter for promote cell proliferation.
