摘要 植物金属硫蛋白在金属离子的储存、运输和代谢及降低重金属离子的毒性等方面起着重要作用。本文研究了Cu2+、Zn2+对丹参毛状根中金属硫蛋白基因表达的影响。以6,7-V为液体培养基,分别以缺乏、正常及原培养基100、500、1000倍的Cu2+、Zn2+处理,采用半定量PCR法检测不同时间点时金属硫蛋基因的表达情况。结果发现Cu2+、Zn2+诱导金属硫蛋基因的表达量分别在100倍处理量时的9h和6d达到峰值。Cu2+、Zn2+能显著诱导丹参金属硫蛋白基因的表达,该基因可能在丹参对铜、锌元素的吸收方面发挥着重要的作用。 Metallothioneins are kind of low-molecular-weight, Cys-rich proteins, which are strongly believed to play important roles in metal metabolism or detoxification. We studied the effects of Cu^2+,Zn^2+ on the expression of Metallothinein gene in hairy root of Salvia miltiorrhiza. Hairy roots of Salvia miltiorrhiza were treated by liquid 6,7-V mediums with default, normal and 100, 500, 1 000 times of Cu^2+,Zn^2+. Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) method was used to moniter the expression level of Metallothinein gene (MT2) in hairy roots of Salvia miltiorrhiza. The results showed that the expression levels of Metallothionein gene achieved the peak value when hairy roots were treated by 100 times of Cu^2+ in 9 h and 100 times of Zn^2+ in 6 d. In this paper we have a primary conclusion that Metallothionein gene might play an important role in absorpting Cu^2+,Zn^2+ in root of Salvia miltiorrhiza based on the evidence of this gene expression induced by Cu^2+,Zn^2+.
