摘要 建立了一个快速、准确、灵敏的液相色谱-串联质谱测定鼠血浆中莪术醇的分析方法.血浆样品经乙腈沉淀蛋白后,以甲醇-乙腈-10mmol/L乙酸铵溶液(体积比:42.5:42.5:15)为流动相,采用反相色谱柱Nueleosil C18分离,通过电喷雾离子化源的四极杆串联质谱仪,以多反应监测方式检测.线性范围为2.5~500.0μg/L,日间和日内精密度〈4.79%,平均提取回收率为100.7%. A rapid, simple and sensitive liquid chromatography/tandem mass spectrometry (LC/MS/MS) method was developed for the determination of eureumol in rat plasma. Samples of the plasma were deproteinated with aeetonitrile, the analytes were then analyzed by reversed-phase HPLC on a Hypersil ODS column with methanol-aeetonitrile-10 mmol/L ammonium acetate (volume ratio: 42.5 : 42.5 : 15 ) as mobile phase. Detection was carried out by multiple reaction monitoring on a API4000 LC/MS/MS system with an eleetrospray ionization (ESI) interface. The assay was linear in a range of 2.5 - 500.0μg/L, intra- and inter-day precisions over this range were 〈 4.79%, the average recovery was 100.7%.
