摘要
根据日本kazusaDNA研究所收录的坛紫菜EST序列AV434021和AV433683设计引物,对坛紫菜配子体的RNA进行RT-PCR扩增,得到产物长度为480 bp。利用DNAstar、Clastal1.81等专业软件进行分析,结果表明,所得产物为紫菜叶绿体的psaA基因片段,其G+C含量为55.9%。G+C含量明显高于A+T的含量;对按核酸序列推导的蛋白序列进行二级结构和三级结构预测,该蛋白存在3个α螺旋和2个β折叠。
The RT-PCR were used for total RNAs from seaweed Porphyra haitanensis as template with primers performed designedon the basis of AV434021 and AV433683 ESTs of the seaweed published by Japanese Kazusa DNA Institute. The eDNA product is 480 bp in length. Analysis by DNAstar, Clastal 1.81 and some professional softwares on website showed that the sequence is homologous to the psaA gene which comprises of 55.9%. There was significantly higher content of G + C than that of A + T. The deduced protein sequence and its secondary and tertiary structure predictions showed that the protein has three α-hellxes and two β-sheets. The results provide some basic information for the study of psaA gene and other related genes of P. haitanensis.
