摘要
目的探讨贲门腺癌(GCA)发生中Ras相关区域家族1A(RASSF1A)基因启动子区异常甲基化的作用。方法采用甲基化特异性PCR法检测81例贲门腺癌及其中20例癌旁正常组织RASSF1A基因启动子区甲基化变化特征,并分析RASSF1A基因启动子区甲基化与肿瘤大体分型、分化程度、淋巴结转移、TNM分期等临床病理特征的关系。结果GCA组织中RASSF1A基因甲基化阳性率(72%,58/81)显著高于癌旁正常组织(5%,1/20)(P<0.05)。GCA组织RASSF1A基因启动子区甲基化与肿瘤大体分型、分化程度、淋巴结转移、TNM分期等临床病理特征无关。结论RASSF1A基因甲基化是GCA癌变过程中频发的分子事件,可能在GCA发生中起一定作用。
Aim: To investigate the significance of RASSF1A gene methylation in gastric cardia adenocarcinoma (GCA). Methods:Methylation-specific PCR was used to analyze the status of RASSF1A gene methylation in 81 cases of GCA and 20 cases of adjacent normal gastric cardiac tissue. Results: The methylation rate of RASSF1A gene in tumor tissue (72% ,58/81 ) was much higher than that (5% , 1/20) in normal adjacent cardiac tissue,P 〈 0. 05. No association was found between the RASSF1A gene methylation and tumor types, differentiation, stages, and metastasis. Conclusion: The RASSF1A methylation is a common event in GCA, which may be related with the carcinogenesis of GCA.
