摘要 目的 通过建立白色念珠菌随机扩增多态性DNA(RAPD)分型技术,结合药敏特性,分析白色念珠菌基因组DNA多态性与耐药性的关系。方法 选择随机引物对受试菌株进行RAPD分析,并优化实验条件。采用GIS2010凝胶成像处理系统对得到的DNA指纹图谱进行分析,生物学软件DNAMAN得出各菌株的遗传距离和亲源关系树状图。结果 筛选出3条随机引物,进行RAPD-PCR;白色念珠菌基因组DNA含量在100ng/μl时,得到的指纹图谱最为清晰;DNAMAN软件分析发现临床分离的5株氟康唑耐药菌株的遗传距离指数较小,其亲源关系也较近。结论 白色念珠菌的基因组DNA多态性与耐药性有密切关系,该结果为白色念球菌的临床诊治及流行病学调查提供了理论依据。 Objective To analyze the relationship between drug resistance and genome DNA polymorphism by establishing the typing technology of random amplified polymorphic DNA and the drug sensitive characteristic in Candia albieans. Methods The high efficient random primers were amplified by RAPD-PCR and the experiment condition of RAPD technique was optimized. DNA fingerprintings were analyzed by GS2010 gel imaging processing system. The genetic distance and genetic relationship tree were obtained in the biological software DNAMAN. Results Three pieces of random primer were selected for RAPD-PCR. The fingerprintings were more clearer when the genome DNA was 100ng/μl. The genetic distance index was little and genetic relationship distance was nearer in the five strains resistant to Fluconazole among 56 C. albicans isolates by DNAMAN. Conclusion There is a close relationship between genome DNA polymorphism and drug resistance in Candida albicans ,which will provide theoretical basis for the clinical diagnosis, treatment and epidemiological investigation.
