摘要
目的研究BCG-CpG-DNA对重组HBsAg的免疫佐剂作用。方法以BCG-CpG-DNA与重组HBsAg混合免疫小鼠,以铝佐剂疫苗为对照,采用放射免疫法检测抗-HBs中和抗体水平,ELISA法分析抗体亚类,酶联免疫斑点试验(ELIS-POT)检测CD8+细胞毒T淋巴细胞(CTL)功能。结果BCG-CpG-DNA能提高抗-HBs中和抗体水平,促进抗原特异性IgG2a的产生,诱导Th1型免疫应答,与铝佐剂协同作用能部分逆转Th2反应。同时能增强抗原特异性CD8+T淋巴细胞的杀伤作用,实验组与对照组比较差异有显著意义。结论BCG-CpG-DNA对HBsAg有较好的免疫佐剂作用,有可能成为一种新型的免疫佐剂。
Objective To study the effect of BCG-CpG-DNA as an immunoadjuvant of recombinant HBsAg. Methods Immunize mice with the mixture of BCG-CpG-DNA and recombinant HBsAg. Determine the anti-HBs neutralizing antibody by RIA, the subclass of antibody by ELISA,the function of CTLs by ELISPOT,and compare with those of HB vaccine containing aluminium hydroxide adjuvant. Results BCG-CpG-DNA increased the anti-HBs neutralizing antibody level, promoted the production of antigen specific IgG2a,induced Thl immune response, and partially reversed Th2 response. The killing activity of antigen specific CD8^+ T lymphocytes of mice in test group was significantly higher than that in control group. Conclusion BCG-CpG-DNA showed good adjuvanticity to recombinant HBsAg, and might be used as a novel immunoadjuvant.
