摘要
目的建立一种基于多元蛋白微阵列的免疫检测方法,用于筛查人血清中艾滋病毒、丙型肝炎病毒。方法将HIV/HCV抗原喷印到活化的玻片表面,抗原与活化基团共价结合后与血清中的特异性抗体反应,用激光芯片扫描系统对蛋白微阵列进行扫描成像。所获得的图像用软件进行分析,并自动判定并生成待测样本的定性结果。结果蛋白微阵列法检测HIV、HCV两种定值血清检测限分别为:12ng/ml、5ng/ml。运用该系统检测HIV、HCV两种抗体参比品,各检测项目的符合率分别为87.5%、93.75%。结论多元蛋白微阵列法特异性强,灵敏度、准确度、精密度较高,具有应用和推广价值。
Objective To establish a immunoassay method that is based on multiplex protein microarray for the screening of HIV, HCV in human serum. Methods Firstly HIV, HCV antigen were printed onto the surface of activated slides. Then the covalent bonding of antigen and activateing group, protein microarray were made up. After these antigens fixed on the surface of slides responsed and combined with special antibodys in serum, protein microarray were scaned and imaged by a high-resolution laser confocal micrarray scanning analysis system. The images were analyzed and assessed by the soft ware,and the qualitative results of these samples were got. Results Detecting HIV, HCV antibodys by protein microarray and comparing with ELISA, the conincidences of all items was respectively 87.5% ,93.75%. Conclusion Multiplex protein microarray has high specificity,sensitivity and accuracy so that deserves extended application.
