摘要 探讨了碱性成纤维细胞生长因子 (bFGF)、谷氨酰胺 ,以及二者联合使用对原代培养的小肠上皮细胞生长和分化的影响。实验分bFGF、谷氨酰胺、bFGF +谷氨酰胺三组 ,于小肠上皮细胞原代培养中加入不同剂量的药物 ,测定细胞的生长和ALP、CK活性。结果显示 :单一bFGF或谷氨酰胺能促进肠上皮细胞生长和减少细胞死亡 ,bFGF使细胞分化降低 ,谷氨酰胺促进细胞分化。二者联合用药的效果明显优于单独用药。 In considering new forms of nutritional support for the epithelial cells of small intestine, we designed this study to investigate basic fibloblast growth factor( bFGF), glutamine,and the combined effects of bFGF and glutamine in vitro on the proliferation and differentiation of small intestinal epithelium,and characterize the interactions between them on the intestinal mucosa. Two groups received bFGF and glutamine at different levels alone. The third group received both bFGF and glutamine at the same levels. Cell proliferation was determined by MTT colorimetric assay. Alkaline phosphatase (ALP) and creatine phosphokinase (CK) activities of epithelial cell were also measured. The data showed that either bFGF or glutamine had a strong trophic effect on the epithelial growth of small intestine in primary culture. The combined effects of bFGF and glutamine on the proliferation of small intestinal epithelium was more than that caused by either agent alone. Furthermore,we found that besides the proliferation, both bFGF and glutamine were able to decrease the appearance of epithelial death. The results were very important for specific nutrients to be chosen in combination to decrease the mucosal atrophy that commonly occurs after intestinal disuse or disease.
