易卒中型肾血管性高血压大鼠血浆NO和TNF-α的动态变化 被引量:5

The Dynamic Changes of Nitric Oxide and Tumor Necrosis Factor-α Levels of Plasma in Stroke-Prone Renovascular Hypertensive Rats

摘要 目的研究易卒中型肾血管性高血压大鼠(RHRSP)血浆中NO和肿瘤坏死因子α-(TNF-α)含量在高血压进程中的动态变化。方法双肾双夹法制作RHRSP模型,分别于术前及术后2、4、8、121、6周舌下静脉取血,硝酸还原酶法测定NO含量,酶联免疫吸附测定法(ELISA)检测TNF-α含量。16周取血后HE染色观察脑组织病理改变。结果RHRSP术后血压持续升高;血浆中NO含量在2周时最高,4周后开始逐渐下降,TNF-α含量逐渐升高,明显异于假手术组;脑动脉血管壁增厚,管腔狭窄。结论高血压大鼠存在血管壁内皮功能异常和炎症反应。血液中NO下降和TNF增高可能是RHRSP出现脑卒中的机制之一。 Objective To observe the activity changes of nitric oxide and tumor necrosis factor- α Levels of Plasma in stroke- prone renovascular hypertensive rats (RHRSP). Methods Two - kidney, two - clip methods were used to induce renovascular hypertension in male Sprague - Dawley rats. Sham- operated rats were used as controls. The levels of serum NO and TNF- α in serum were measured by nitrate reductase and enzyme - linked immunosorbant assay(ELISA) ,respectively, at pre - operation, and 2 w, 4 w, 8 w, 12 w, 16 w after operation.Then brain sections stained with hematoxylin- eosin (HE ) were examined under a microscope. Results Blood pressures keep on increasing after operation in the RHRSP; the Contents of NO in the RHRSP reached peak at 2 w after operation, then decreased from 4 w postoperation , the contents of TNF - α were increased markedly as compared with Sham - operated group. HE - stained cerebrovascular showed enhanced mural thickness and stenotic lumen. Conclusions These fingdings suggest that vessel wall endothelial dysfunction and vascular inflammation exist in hypertensive rats. Down - regulative activity of NO and up - regulative activity of TNF - α may play a role in the onset of stroke in the RHRSP.
