摘要
目的比较替米沙坦与罗格列酮对胰岛素抵抗(IR)大鼠的血糖、血脂、及血胰岛素水平的影响。方法采用高果糖饲料喂养雄性SD大鼠4周建立IR大鼠模型,并通过检测相应的指标判断模型是否成功。SD大鼠随机分为:对照组I、R组、IR+替米沙坦组、IR+罗格列酮组。每两周检测各组大鼠的尾动脉收缩压。第8周末采血检测空腹血糖、血脂、血胰岛素水平,并计算胰岛素敏感指数(ISI)。结果(1)与IR组相比,IR+替米沙坦组、IR+罗格列酮组大鼠的收缩压下降(P<0.01),空腹血糖、血胰岛素、甘油三酯、总胆固醇浓度也显著降低(P<0.01),ISI则明显提高(P<0.01)。(2)IR+替米沙坦组与IR+罗格列酮组相比,空腹血糖、血胰岛素、甘油三酯、总胆固醇浓度及ISI间差异无显著性。结论替米沙坦与罗格列酮都能提高胰岛素抵抗大鼠的ISI,改善IR,纠正糖脂代谢的紊乱,且两者的作用差异无显著性。
Objective To compare the effect of telmisartan and msiglitazone on sugar and lipid metabolism in insulin resistance (IR) rats. Methods Male SD rats were fed with high fructose diet for 4 weeks to establish IR rats model. To verify IR rats model, the following indexes were measured respectively in each group. SD rats were randomly divided into control, IR, IR + telmisartan and IR + msiglitazone group. Systolic blood pressure( SBP)was measured every two weeks, fasting blood sugar(FBS), blood lipid and the level of fasting serum insulin(FSI)were measured respectively in groups at the end of week 8, and insulin sensitive index(ISI) was calculated. Results ( 1 ) Compared with the IR group, SBP, FBS, triglyceride, blood total cholesterol and the level of FSI were significandy lowered ( P 〈 0.01 ) ; ISI was significantly increased in the IR + telmisartan group and IR + rosiglitazone group (P 〈 0.01) (2)FBS, triglyceride, blood total cholesterol, the level of FSI and ISI were not significantly different between the IR + telmisartan group and IR + rosiglitazone group. Conclusion Telmisartan and rosiglitazone can increase ISI and improve insulin resistance, correct the abnormity of sugar and lipid metabolism in IR rats.
