摘要
目的研究二烯丙基二硫(DADS)诱导人髓性白血病HL-60细胞分化过程中蛋白质组的差异表达。方法运用蛋白质组学方法(双向凝胶电泳、质谱分析、生物信息学方法)分析鉴定DADS诱导HL-60细胞分化的差异表达蛋白质。结果DADS作用HL-60细胞后,细胞形态发生明显变化,NBT还原能力明显增强。与未处理组比较,初步鉴定的差异蛋白质DJ-1蛋白在处理组中表达下调,其功能与转录调节有关。结论DADS可能通过抑制DJ-1蛋白合成,活化PIAS蛋白抑制STATs转录因子的活性,进而抑制JAKs/STATs信号传导通路,诱导HL-60细胞分化。
Objective To study differential proteomic expression in the differentiation of HL - 60 cells induced by diallyl disulfide (DADS). Methods Differential expression of cellular proteins in HL - 60 cell induced by DADS were analyzed and identified via proteomic approach, two dimensional gel electrophoresis, mass spectroscopy and bioinformatics. Results The cytomorphosis of the DADS - treated cells was significantly different from controls. NBT reduction was increased in the DADS- treated cells. Among the identified differential expression protein, the expression of DJ- 1 protein was decreased in DADS- treated group. Its function was associated with transcription. Conclusions Inhibition expression of DJ - 1 protein was involved in HL- 60 cells differentiation induced by DADS and might associate with inhibition of JAKs/STATs single pathways via activating PIAS protein.
