摘要
目的研究1.0、2.0和3.0μmol/L的三氧化二砷(As2O3)对人宫颈癌Hela细胞体外生长的影响,并初步探讨其机制。方法应用1.0、2.0和3.0μmol/L As2O3作用于人宫颈癌Hela细胞系,采用MTT比色法检测细胞生长情况;运用流式细胞术检测细胞凋亡率;应用免疫组化SABC法检测细胞中Bcl-2和Bax蛋白的表达。结果As2O3能抑制宫颈癌Hela细胞的体外生长,诱导宫颈癌Hela细胞凋亡,并呈浓度和时间依赖性(P<0.05)。As2O3能引起细胞内Bcl-2蛋白表达下调和Bax蛋白表达上调(P<0.05)。结论As2O3对宫颈癌Hela细胞体外生长具有明显抑制作用,并诱导宫颈癌Hela细胞凋亡,可能与其引起细胞内Bcl-2蛋白表达下调和Bax蛋白表达上调有关。
Objective To study the effect induced by 1.0,2.0,3.0 μmol/L arsenic trioxide(As2O3 ) on human cervical carcinoma Hela cell lines. Methods Hela cell lines were treated by 1.0,2.0,3.0 μmol/L As2O3. Cell proliferation was evaluated by MTT assay. Flow cytometry was used to examine apoptosis on cervical carcinoma Hela cell lines. The expression of Bcl - 2 protein and Bax protein were investigated by immunohistochemical SABC techniques. Results MTT assay showed that As2O3 inhibited human cervical carcinoma Hela cell lines growth in vitro in a dose and time - dependent manner. Flow cytometry showed that As2O3 induced the apoptosis in a dose and time - dependent manner ( P 〈 0.05 ). Immunohistochemical SABC techniques showed that As2O3 can downregulate the expression of Bcl - 2 protein and upregulate the expression of Bax protein( P 〈 0.05 ). Conclusions As2O3 can significantly inhibit Hela cells growth and proliferation and induce their apoptosis. The reason of these changes may be the downregulation expression of Bcl - 2 protein and upregulation expression of Bax protein.
