摘要
目的探讨N-乙酰半胱氨酸(NAC)对博莱霉素所致大鼠肺纤维化血清MDA的影响。方法取健康、雌性Wistar大鼠75只,随机分为三组:博莱霉素模型组(B组);NAC治疗组(N组):模型制备同B组,于博莱霉素处理前1周及此后每日经强饲法口服NAC(3mmol/kg);正常对照组(C组):同样条件下向气管内注入等量生理盐水。三组动物分别于气管灌注后的第1、3、7、14及28天随机处死五只。取肺组织标本行病理学观察、肺系数测定及血清丙二醛(MDA)检测。结果肺系数N组与B组统计学分析显示除第1天外其余各时间点差异均有显著性(P<0.05);B组与C组比较,第3、7、14、28天MDA升高(P<0.05),而N组与C组相比,第1、14、28天MDA差异无显著性(P>0.05),第3、7天差异存在显著性(P<0.05)。B组与N组血清MDA含量均第7天达到高峰,而后逐渐下降,进行两组间统计学比较,第7、14、28天差异存在显著性(P<0.05)。结论NAC可降低博莱霉素所致肺纤维化大鼠MDA的产生,通过抗氧化作用抑制肺纤维化的发展,对肺纤维化具有较明显的保护作用。
[Objective] To investigate the interventional effect of N-acetylcysteine on blood serum MDA of pulmonary fibrosis rat model. [Methods] Seventy-five female rats were randomly categorized into bleomycin group(B group), N-acetylcysteine-treated group(N group) and control group (C group). Except the normal control group ,other rats received instilling bleomycin. B group was fed on NAC (3 mmol/d/kg) before treated by bleomycin a week ago and C group was instilled by normal saline instead of bleomycin. Five rats in each group were sacrificed at days 1, 3, 7, 14 and 28 after intratracbeal instillation. Histopathological examination, the lung coefficient and the level of MDA in blood serum were observed. [Results] Firstly, The lung coefficient in N group was remarkably different from that of B group except at day 1. Secondly, The level of MDA in blood serum of B group was significantly higher than that of C group at days 3, 7, 14, 28. (P 〈0.05). N group with C group had significantly difference at days 3, 7 (P 〈0.05). Furthermore, B group and N group had the same change tendency, and the MDA of two groups all reached the peak at days 7, then gradually reduced, but two groups had remarkably difference at days 7, 14, 28. [Conclusion] The MDA in bleomycin-induced pulmonary fibrosis in rat can be reduced by NAC, and anti-oxidation means can inhibit the development of lung fibrosis, which will be a new ways to treat lung fibrosis.
