摘要
目的:研究肺癌穿刺标本中Survivin mRNA的定量表达情况。方法:用Trizol方法提取肺穿刺组织(54例)总RNA后,将其逆转录为cDNA,建立实时荧光定量RT-PCR方法,测定肺穿刺标本组织Survivin mRNA的表达。结果:(1)Sur-vivin mRNA在肺癌中高度表达(0.26±0.023)logcopies/μg(阳性率为91%),与良性肺病变中表达(0.05±0.009)log copies/μg(阳性率为12.5%)均有显著性差异(P<0.01);(2)肺癌穿刺敏感性、Survivin表达阳性率均为91%(42/46),细胞病理学与Survivin检测联合诊断,肺癌的检出率达100%。结论:运用RT-PCR法检测肺癌穿刺标本Survivin mRNA的表达,对提高肺癌的诊断有一定的帮助。
Objective: To investigate the expression of survivin mRNA in percutaneous transthoracic needle aspiration (PTNA) specimens with a RT-PCR. Methods:Total RNA was extracted with Trizol from the tissues of lung lesions and mRNA was transcribed reversely into eDNA. The real-time quantitative RT-PCR was used to detect the expression of survivin mRNA. Results: ( 1 )The expression of survivin mRNA in the lung carcinoma tissues as (0. 26 ±0. 023) log copies/μg (the positive rate as 91%o, 42/46) was significantly higher than that in the benign lesions tissue as (0. 05 ±0. 009) log copies/μg (the positive rate as 91%, 1/8) (P 〈 0. 01 ). (2) The sensitivity of survivin mRNA detection combined with histology in PTNA specimens increased ( as 100% ) , was higher than that of histology examination or survivin mRNA alone (as 91% ) respectively. Conclusion:Real-time fluorescence quantitative RT-PCR can be used to detect the expression of survivin mRNA in PTNA specimens. Detecting survivin mRNA may be used in diagnosis of lung cancer.
