摘要 RNA干扰作用(RNA interference,RNAi)是由双链RNA(double-stranded RNA,dsRNA)引发的转录后基因沉默机制(post transcriptional gene silencing,PTGs)。RNAi主要是指dsRNA分子进入细胞内,特异性降解与之同源的mRNA,从而特异高效的抑制相应基因表达活性的现象。它不但是研究基因功能的有力工具,而且为特异性基因治疗提供了新的技术手段。本文介绍RNAi的作用机制与应用的研究进展。 RNA interference (RNAi) is a post-transcriptional gene silencing induced by introduction of double-stranded RNA (dsRNA). It's a process of sequence-specific gene silence,which can specifically and effectively suppress the activity of corresponding mRNAs induced by double-stranded dsRNA. This is a highly specifically mechanism that involved in kinds of proteins. It's not only be a powerful tool in gene functional research, but also provides a new technique for gene therapy. RNAi not only is a powerful method for studying genes,but also offers new techniques for the therapy of specific gene.This paper introduces the research progress in the mechanisms and appfication of RNAi.
