摘要
目的:观察冠心病患者循环内皮祖细胞数量与分化相关功能的变化特点,探讨体外条件下不同浓度阿托伐他汀对冠心病患者循环内皮祖细胞数量和增值,黏附,迁移等分化相关功能的影响。方法:以16例条件齐同的非冠心病患者作对照,观察16例冠心病患者循环内皮祖细胞数量与功能的变化,体外条件下不同浓度阿托伐他汀处理冠心病患者循环内皮祖细胞。结果:冠心病患者外周血内皮祖细胞的数量与功能下降,阿托伐他汀呈浓度依赖性提高冠心病患者内皮祖细胞的数量与增值,黏附,迁移等分化相关功能;阿托伐他汀对内皮祖细胞分化相关功能的改善作用大于对其数量的影响。结论:阿托伐他汀可上调冠心病患者循环内皮祖细胞的数量与分化相关功能,且以改善功能为主。
Objective: To investigate the effects of atorvastafin with different concentrations on the feature of the quantity and differentiation-correlated functions like proliferation, adhesion and migration of circulating endothelial progenitor cells isolated from patients with coronary heart disease, Method: 16 patients with coronary heart disease (CHD) and 16 matched control subjects were enrolled, Total mononuclear cells were isolated from.peripheral blood by density gradient centrifugation, and then the cells were plated on gelatin-coated culture bottles. After cultured for 7 days, attached cells were stimulated with atorvastafin in series of final concentration: 0.1μmol/L, 1μmol/L and 10μmol/L, EPCs were characterized as adherent cells not only double positive for Dil-acLDL-uptaking and lecfin-binding, but also double expressive of VEGFR2 and AC133 by laser scanning confocal microscope with direct fluorescent staining. EPCs proliferation and migration were measured by MTT assay and modified by Boyden chamber assay, respectively, EPCs adhesion assay was performed by counting the adherent cells number when EPCs were replated on gelatin-coated dishes. Results: The number and proliferating, adhering, migrating function of EPCs, which are related with cell differentiation, were significantly reduced in patients with CHD, compared with the control subjects. Treated with atorvastafin, EPCs' number and proliferating, adhering, migrating function were significantly improved. Especially, when treated with high dosage of atorvastatin (10μmol/L final concentration), EPCs' adhering and migrating function recovered to the normal level, surpassing the improvement in number and proliferating. Conclusion: Atorvastafin can upgrade the number and differentiation-correlated functions, including proliferation, adhesion and migration of EPCs isolated from patients with coronary heart disease. It mainly improved the functions
