摘要
目的探讨不同程度缺血再灌注损伤脊髓的病理变化特征及其意义。方法40只成年家兔随机均分为假手术组、缺血30min组、缺血45min组和缺血60min组,以肾下腹主动脉阻断法制备脊髓缺血再灌注模型,术后2 d时,采用免疫组织化学法和苏木精-伊红染色观察腰髓病理学改变。结果缺血时间越长,再灌注脊髓病理损伤程度越重,伤后2 d时可见受损脊髓发生出血、水肿、变性坏死,白细胞浸润以及NF-κBp65I、CAM-1表达增加。结论该模型具有重复性好,能较好地反映分级缺血再灌注损伤脊髓的病理变化特点。
Objective To investigate the pathological characters of spinal cord after graded ischemia/reperfusion injury and its significance. Methods The model of ischemic and reperfusional irjury of spinal cord was created by clamping the infrarenal aortic in rabbits 40 adult rabbits, which were randomly divided into sham-operation group.ischemia for 30min,45min and 60min group. After 2 days of reperfusion, the pathological changes of lumbar segmental spinal cord were observed with the HE staining and immunohistologicalmethnd. Results The longer the ischemic time, the more severe the pathological damage of spinal cord tissue after reperfusional injury. Hemorrhage, swelling, degeneration of the lumbar spinal cord tissue, the remarkable expression of ICAM-1. NF-κBp 65 and leukocytes infiltration occurred in reperfusion 48 hours. Conclusion This model was reproducible for the pathological charaters graded spinal cord isehemia/reperfusion injury eompard with other models.
