微孔板比色测定法在微量蛋白测定中的意义 被引量:1

摘要 目的建立一种准确、快速的微克级蛋白质测定方法。方法以考马斯亮蓝作为显色剂,用酶标仪微孔板比色法与常规Bradford法、紫外吸收法、改良Lowry法同时测定99例样本蛋白质含量,并比较检测结果的相关性。结果改良微孔板Bradford法与常规Bradford法样品的测定结果相近,两者有良好的相关性(r=0.991,P〈0.001)。结论微孔板Bradford法是一种简单、快速、灵敏的蛋白质测定方法,尤其适用于大批量样品、微量样品的蛋白质含量测定。 Objective To establish an accurate and rapid detective method for microgram grade proteins. Methods Coomassie Brilliant Blue was used as color reagent, and protein levels in 99 samples were detected simultaneously and respectively by micro-well colorimetry (using enzyme labeler), common Bradford assay, ultraviolet absorbant assay and improved Lowry's method. Then, correlation of the results were analyzed. Results Results of improved micro-well plate Bradford assay were similar to that of common Bradford assay, and the correlation between them was significant (r= 0. 991, P〈 0. 001). Conclusion Micro-well plate Bradford assay is a simple, rapid and sensitive method for protein detection, especially for large batch and trace protein detection.
