附子种质资源HPLC指纹图谱的初步研究 被引量:4

摘要 为构建生附子药材的HPLC品质指纹图谱,建立附子种质资源品质评价高效准确的鉴定方法,以收集自四川、云南等地的14份附子种质资源和四川江油附子GAP基地的参照药材为材料,采用HPLC方法,在kromasil C18柱,水(6 mmol·L-1SDS、20mmol.L-1NaH2PO4)-甲醇,梯度洗脱,流速0.8 mL·min-1,检测波长240 nm等条件下建立生附子品质指纹图谱。结果表明各份供试样品色谱峰分离良好,符合指纹图谱有关规定,15份供试样品有9个共有峰,与参照药材比较相似度0.90以上的有7批。本研究所构建的指纹图谱可作为生附子药材鉴定和质量控制的参考依据,建立的HPLC方法可作为附子良种选育研究中对种质资源内在品质进行鉴定的有效方法。 To construct the fingerprint by HPLC and develop a high-efficient and concise method for medicinal quality evaluation of Radix Aconiiti Lateralis Preparata. Fourteen germplasms from Sichuan and Yunnan, one criterion from Jiangyou, Sichuan were sampled. The quality fingerprint were constructed by HPLC under the chromatographic condition, such as,the column :kromasil C18, the mobile phase:water(6 mmol · L ^-1 SDS,20 mmol · L^-1 NaH2PO4 )-methanol,the flow rate:0.8 mL · min^-2 and the detection wavelength:240 nm. The chromatogram showed that the peaks were separated and accorded with the fingerprint of the criterion. Among 15 samples 9 shared peaks were indentified. There were 7 samples whose similarities with the criterion were above 0.90. In conclusion, the constructed fingerprint can be used as the reference for medicinal quality evaluation of Radix Aconiiti Lateralis Preparata. HPLC can be used as a high-efficient method to evaluate the quality of gennplasms for breeding of Radix Aconiiti Lateralis Preparata.
