摘要
[目的]建立一个HPLC方法,对国内13个不同产地柴胡样品中的柴胡皂苷a、d进行含量测定。[方法]色谱柱:YMC-Pack ODS-AC18(250×46mm,5μm),流动相乙腈-水(43∶57),检测波长:203nm。[结果]柴胡皂苷a、d的线性回归方程分别为Ya=1·223×106X-2·2498×105(r=0·9999)、Yd=2·093×106X-4·7862×105(r=0·9998),平均加样回收率分别为98·13%、97·99%。[结论]本研究建立的柴胡皂苷a、d含量测定方法简便易行,结果准确、可靠,便于对柴胡药材进行质量控制。
[Objective]To develop a method using HPLC for determination content of saikosaponin a and saikosaponin d of Radix Bupleuri from different areas. [Method]Column: YMC-Pack ODS-A C18 (250 × 46 mm, 5μm) , mobile phase: acetonitrile-water(43 : 57),detected wavelength:203nm. [Results]The regression eguations of saikosaponin a and saikosaponin d were Ya= 1. 223 × 10^6 X - 2. 249 8× 10^5 (r =0. 999 9) ,Yd = 2. 093× 10^6 X - 4. 786 2× 10^3 (r =0. 999 8). The average recovery rates of saikosaponin a and saikosaponin d were 98.13 %,97.99 %. [Conclusion]The HPLC method is simply and suitable to control quantitative of radix bupleurum.
