摘要
在使用10mmol/L NH4Ac(pH 4.5),30mmol/L Cu(Ac)2和60mmol/LL-羟基脯氨酸的优化条件下,13min内拆分了3种α-羟基酸对映体(DL-苹果酸、DL-苦杏仁酸和DL-对溴苦杏仁酸),除苹果酸外均获得了满意的拆分结果;考察了电泳缓冲液组成、pH值等影响分离效果的因素。
A rapid and simple approach to chiral separation of α-hydroxy acids by ligand exchange capillary zone electrophoresis is described in this paper. A Cu ( Ⅱ) - L-hydroxyproline complex was used as chiral selector. Its applicability was demonstrated with α-hydroxy acids, namely D, L-malic acid, D,L-mandelic acid and D,L-p-bromomandelic acid. The optimal resolution of mixed α-hydroxy acids was obtained with a running buffer solution consisted of 10 mmol/L NH4Ac( pH 4. 5 ), 2 : 1 concentration ratio of L-hydroxyproline to Cu ( Ⅱ), 30 mmol/L Cu (Ac)2, and 60 mmol/L L-hydroxyproline. The influences of pH and composition of background buffer solution on the resolutions were investigated. Using the proposed method, three different α-hydroxy acids except D,L-malic acid were separated successfully in less than 13 min simultaneously.
