摘要 目的:应用荧光定量RT-PCR(Fluorogenic quantitative reverse transcriptase polymerase chain reaction,RT-PCR)检测肺癌外周血中的表达情况,并探讨其表达的临床意义。方法:用荧光定量RT-PCR方法检测67例肺癌患者外周血CK19 mRNA表达。结果:66例肺癌患者外周血中CK19mRNA检测阳性35例,阳性率53.0%,统计结果显示外周血CK19mRNA的表达与性别、年龄、病理类别、临床分期无关,在不同分化程度、淋巴结转移状态间有显著差异。结论:CK19mRNA可作为RT-PCR法检测肺癌患者外周血微转移的分子标志物,有助于早期诊断肺癌血行转移,指导临床治疗。 Objective:To investigate the CK19 mRNA expression in peripheral blood and its significance in lung cancer patients by fluorogenic quantitative reverse transcriptase -polymerase chain reaction( RT- PCR). Methods: CK19mRNA expression in peripheral blood was detected in 66 cases of lung cancer by RT - PC1L Results: Positive CK19mRNA was detected in peripheral blood in 35 of 66 patients with lung cancer. The positive rate was 53.0%. Statistical analysis indicated that expression of CK19mRNA in peripheral blood was not correlated with gender, age, pathological type of the cancer and clinical TNM stage, but had significant difference in different cell differentiation, lymphy node metastasis status. Conclusion: RT - PCR amplification of CK19mRNA is a method to detect early hematogenous dissemination of cancer cells for patient with lung cancer,and may lead to early diagnosis of patients with subclinical metastasis in circulation.
