摘要
对影响分光光度计法测定果胶酶活力的因素进行了研究,结果表明,不同的测量波长、DNS试剂用量、果胶用量和果胶酶稀释倍数对果胶酶活力测定结果都有显著影响。最终确定适宜的波长为540nm、DNS试剂(3g/L)用量为2.5mL、果胶(5g/L)用量为1mL、酶液稀释倍数为1000倍。
The factors influencing result of pectinase activity analysis by spectrophotometry were investigated here. The results showed that the wavelength, the dosage of DNS and pectin, and the dilution of pectinase all remarkable effected on the result. The optimal measuring condition is wavelength at 540nm, DNS (3g/L) 2.5mL, pectin (5g/L) 1mL, pectinase 0.1%.
