口腔鳞状细胞癌间质成纤维细胞α-SMA的表达及意义 被引量:5

摘要 目的探讨口腔鳞状细胞癌(OSCC)间质成纤维细胞中α-平滑肌肌动蛋白(α-SMA)的表达水平及意义。方法收集新鲜OSCC组织标本34例,以正常口腔黏膜(5例)、口腔扁平苔藓(6例)和口腔白斑组织(9例)为对照,在冰冻切片上行α-SMA和波形蛋白免疫组化染色(SP法)。结果OSCC组α-SMA的平均染色分值和染色阳性标本百分率均高于对照组(P<0.01);在OSCC组,随着细胞分化程度的逐渐降低,α-SMA平均染色分值和染色阳性标本百分率均有逐渐升高的趋势(P<0.05);在α-SMA染色阳性区域波形蛋白染色亦呈阳性。结论α-SMA在癌间质成纤维细胞中的表达可作为OSCC的辅助诊断及严重程度的判定指标。 Objective To study the expression of α-SMA in stromal fibroblasts of different pathological grades of OSCC. Methods Fresh specimen of 34 cases of OSCC and 20 cases of controls (including 5 cases of normal oral mucosa, 9 cases of oral leukoplakia and 6 cases of oral lichen planus) were collected and made into freezing slides. The immunohistochemistry was used to detect the expression of α-SMA and vimentin. Results The mean percentage of positive cases and staining score of α-SMA in OSCC group were higher than control group with statistical significance (P〈0.01). Percentage of positive cases and staining score of α-SMA were increased with the decreased level of cell differentiation in OSCC group (P〈0.05). The α-SMA positive cells also showed positive for vimentin, which implied to be carcinoma-associated fibroblasts with smooth muscle phenotype transformation. Conclusion α-SMA expression in stromal fibroblasts of OSCC could be used as an index of OSCC in its supplementary diagnosis and severity judgement.
