胃癌组织中KAI1 mRNA表达的意义

Expression and clinical significance of KAI1 mRNA in gastric carcinoma

目的探讨KAI1基因和蛋白在正常胃黏膜、不典型增生胃黏膜及癌组织中的表达及其与之发生、发展的关系.方法应用原位杂交技术和免疫组织化学技术检测不典型增生胃黏膜65例及胃癌组织74例中KAI1 mRNA和KAI1蛋白的表达.结果KAI1 mRNA和KAI1蛋白在胃癌组织中表达显著降低,与正常胃黏膜、不典型增生胃黏膜组织中表达的差异有显著性意义(χ2=36.328,P<0.05).在胃癌组中不同的浸润深度、有无淋巴结转移和脉管侵犯组中阳性表达率的差异性有统计学意义(P<0.05);而在年龄、性别组间的差异性无统计学意义(P>0.05).结论KAI1 mRNA和蛋白低表达在胃癌组织中的发生、发展中可能起着重要作用,有望成为早期诊断、评估肿瘤细胞侵袭转移潜能的指标之一.

AIM： To study the expression of the metastasis suppressor gene KAI1 in gastric tumors and the relationship between the expression of KAI1 and the tumor genesis and development. METHODS： The expression of KAI1 mRNA and protein was investigated with in situ hybridization and immunohistochemistry （SP method） respectively in normal gastric tissues （n = 22）, tumor tissues（ n = 74） and tissues of atypical gastric mucosal hyperplasia（n = 65）. RESULTS ： The positive expression of KAI1 mRNA and protein in gastric carcinoma was significantly reduced and showed statistical difference as compared with normal gastric mucosa and atypical hyperplasia （x^2 = 36. 328, P 〈 0.05）. The expression of KAI1 mRNA and protein in gastric carcinoma was correlated to the depth of infiltration of carcinoma, vessel invasion and lymph node metastasis （P 〉 0.05）, but not to with patient＇s age and gender （ P 〉 0.05 ）. CONCLUSION： KAI1 may play an important role in tumor genesis, development, invasion and metastasis of gastric carcinoma, and can serve as a molecular marker for early diagnosis and prediction of invasion and metastasis of gastric carcinoma.
