摘要
目的制备小鼠抗人源性BART的抗体。方法采用原核系统表达人全长BART,经纯化后免疫小鼠制备抗BART的抗体,以Western blot、免疫组化染色法检测抗体的效价和特异性。结果获得高效价的小鼠抗BART的抗体,该抗体能够识别大鼠脊髓组织中的BART分子。海马神经元和星形胶质细胞的免疫组化染色结果显示,BART分子在海马神经元、星形胶质细胞中广泛表达;在海马神经元中,BART与钙离子通道存在共定位现象。结论制备出能特异性识别天然BART分子的抗体,为检测BART分子并进一步研究其功能奠定了基础。
AIM: To prepare mouse anti-human BART polyclonal antibody (Ab) and characterize its properties. METHODS: BART protein was expressed in E. coil BL21. Mouses were immunized with purified BART protein to prepare anti-BART Ab. The titer and specificity of the prepared antibodies were identified by Western blot and immunohistochemical staining, respectively. RESULTS: Purified mouse anti-human BART Ab with high titer was obtained. The anti-BART Ab showed good specificity and could be used to detect the BART in rat spinal cord tissues. Immunohistochemical staining indicated that BART was expressed widely in hippocampus neurons and astrocytes. BART and calcium ion channel were colocalization in hippocampus neurons. CONCLUSION: Successful preparation of anti-human BART antibody provides a useful tool for identification and further functional study of BART molecule.
