摘要
建立了测定枸杞多糖各级分中半乳糖醛酸含量的高效液相色谱法。枸杞多糖各级分中半乳糖醛酸含量用Sugar-Pak Ⅰ柱进行定量分析。流动相为0.0001mol/L EDTA-Ca,流速为0.5ml/min,以折光检测器检测,平均回收率为97.7%(n=3),变异系数0.2985%(n=5),进样量在0~28μg范围内线性关系良好,r=0.9990。该方法比常规的硫酸-咔唑法快速、简便,分析结果准确可靠。
A high perfomance liquid chromatograph method for the determination of Dgalacturonic acid in Lycium chinense polysaccharide (LCP) was estabolished. The polysaccharide was extracted from water after removing impurities and then separated on column packed with EDAE-cellulose. The four fractions of LCP, namely LCP-Ⅰ, LCP-Ⅱ, LCP-Ⅲ and LCP-Ⅳ were obtained by water-salt as eluant. The quantitative analysis of D-galacturonic acid in all of fractions were performed by HPLC using a sugar-park Ⅰ column with 0. 0001 mol/L EDTA-Ca in water as eluant and monitored by RID. The calibration curve was linear in the range of 0~28 μg of D-galacturonie acid with correlation coefficient of 0.9990. The varied coefficient and average recovery rate were 0.2985 % (n=5) and 97.7 % (n=3) respectively.
