摘要
研究了实验室筛选出的高产油脂酵母菌(Candida Yeast-1-1)产脂的最佳条件,在此基础上,通过多因素正交试验得出尿素包合法提取酵母油脂中不饱和脂肪酸的最佳方法。试验结果表明,在温度为30℃,摇床转速120r/min的条件下,酵母菌产脂的最佳培养基为:0.02%的MnCl_2、0.03%的FeSO_4、0.3%的(NH_4)_2SO_4、3%的葡萄糖、pH6.5;油脂提取的最佳尿素包合条件为:尿素和脂肪酸的比例为3:1,回流时间30min,冷却析出温度0℃。
The fat-producing optimum conditions of high-yield yeast(Cand/da Yeast -1-1) were studied in this experiment. An optimum method of yeast fat urea inclusion complex of saturated fatty acids using multi-criteria cross experiment was obtained. The optimized liquid fermentation mediums are as follows: 0.02% MnCh, 0.03% FeSO4, 0.3% (NH4)2SO4, 3%C6H60 and pH6.5, at 120 r/rain and 30℃. The optimum exwacting conditions of yeast fat urea inclusion complex arc: ratio of urea and fatty acids(3:1), refluxing time (30 rain) and cooling temperature (0 ℃).
