目的:探讨干细胞因子(stemcellfactor,SCF)与卵泡发育及妊娠之间的关系。方法:45例因输卵管因素接受体外受精-胚胎移植(invitrofertilizationandembryotransfer,IVF-ET)的不孕患者,在超促排卵后取卵的同时留取卵泡液和血清标本。用酶联免疫分析法(ELISA)测定血清和卵泡液中的SCF水平,用放射免疫分析法(RIA)测定雌二醇(E2)、孕酮(P)和睾酮(T)水平。结果:妊娠组(n=16)血清和卵泡液中SCF水平均高于未妊娠组(n=29),差异有统计学意义(P<0.05),E2水平和E2/T、E2/P比值也高于未妊娠组(P<0.05);卵泡液中P水平显著低于未妊娠组(P<0.05),2组T水平差异无统计学意义(P>0.05)。血清与卵泡液中的SCF与E2水平呈正相关(r=0.56,t=4.44,P<0.01;r=0.32,t=2.22,P<0.05),与P和T水平均无直线相关性,卵泡液中的SCF与E2/T(r=0.40,t=2.86,P<0.01)和E2/P(r=0.37,t=2.64,P<0.05)比值呈正相关。结论:SCF可能对卵泡发育和妊娠过程有促进作用。
Objective: To investigate the relationship of stem cell factor (SCF) levels in serum and follicular fluid with follicular development and pregnancy. Methods: Forty-five infertile patients received in vitro fertilization and embryo transfer (IVF-ET) in our hospital, and all patients underwent a standard leuteal phase started with gonadotropic hormone-releasing-agonist (GnRHa) according to the long cycle protocol of ovulation hyperstimulation. At the time of transvaginal oocyte retrieval, serum samples were obtained from peripheral vein. Follicular fluid (FF) obtained from mature oocyte-bearing follicles. SCF levels in serum and FF were measured in duplicate by a solid phase enzyme-linked immunosorbant assay (ELISA). Estradiol (E2), progesterone (P) and testosterone (T) levels were measured by radioimmunoassay (RIA). Results: Concentrations of SCF and E2 and ratios of E2/T and E2/P in serum and FF were significantly higher in pregnant patients (n=16) than those in nonpregnant patients (n=29). The level of P in FF was significantly lower in pregnant patients than that in non-pregnant patients (P 〈 0.05). No significant differences of T levels in serum and FF were found between pregnant patients and non-pregnant patients. Positive correlations were found between the levels of SCF and E2 in serum (r = 0.56,t = 4.44, P 〈0.01) and FF (r = 0.32, t = 2.22, P 〈 0.05). No positive correlations were found between the levels of SCF and the levels of P or T in serum and FF. Additionally, the positive correlations were found between the levels of SCF and E2/T (r = 0.40, t = 2.86, P 〈 0.01) or E2/P(r = 0.37, t = 2.64, P 〈 0.05)in FF. Conclusion: SCF may play a role in follicular development and participate the pregnant process.
