胶质瘤中hTERT、VEGF表达及其与肿瘤血管形成的关系 被引量:2

Expression of hTERT and VEGF in gliomas and its relation to tumor angiogenesis

摘要 目的探讨胶质瘤中端粒酶逆转录酶(hTERT)、血管内皮生长因子(VEGF)的表达与肿瘤血管形成间的相互关系。方法分别用原位杂交和免疫组化染色检测106例胶质瘤hTERT和VEGF的表达;用CD34标记瘤组织血管内皮细胞,测定微血管密度(MVD)。结果hTERT、VEGF总阳性表达率分别为53.8%(57/106)、68.0%(72/106);hTERT或VEGF阳性分别定位于肿瘤细胞核内和细胞质内。hTERT阳性或VEGF阳性的瘤组织MVD分别为71.2±18.0和74.4±20.0;而相应的阴性组分别为60.3±21.8和58.4±23.1,两组差异均有显著性(P<0.01)。hTERT、VEGF的表达和MVD均与胶质瘤组织病理分级呈正相关(P<0.01)。结论hTERT、VEGF的表达及MVD均与胶质瘤的恶性程度有关,前两者阳性表达,其MVD高于两者阴性表达,说明hTERT、VEGF在肿瘤血管形成中可能起促进作用。 Purpose To study the relation of hTERT and VEGF expression to tumor angiogenesis in gliomas. Methods In situ hybridization (ISH) and immunohistochemical staining were used respectively by detecting expression of hTERT and VEGF in 106 cases of gliomas. The endothelial cells of blood vessels within the tumor were labeled by CD34 for calculating MVD. Results Total positive expression rates of hTERT and VEGF were 53.8% (57/106) and 68.0% (72/106) ; hTERT and VEGF expression located in the nucleus and cytoplasm of tumor cells; MVD in hTERT-positive and VEGF-positive group was 71.2 ± 18. 0 and 74. 4 ±20. 0, but MVD in hTERT-negative and VEGF-negative group was 60. 3 ±21.8 and 58. 4 ± 23. 1, respectively. MVD was significantly higher in the hTERT-positive or VEGF-positive gliomas than in the negative ones (P 〈0.01 ). hTERT, VEGF expression and MVD were all positively correlated with histopathological grade of glioma ( P 〈 0. 01 ). Conclusions Expression of hTERT, VEGF and MVD is all related to malignant degrees in gliomas; MVD in the hTERT-positive or VEGF-positive gliomas is higher than the negative ones, suggesting that hTERT and VEGF may play promoting roles in tumor angiogenesis.
