摘要
目的:研究白背叶的化学成分并建立含量测定方法。方法:聚酰胺柱色谱,Sephadex LH-20柱色谱,制备液相色谱分离纯化,理化性质和光谱数据进行结构鉴定;高效液相色谱条件,Inertsil ODS-3 C18(4.6 mm×150mm,5μm)色谱柱,流动相为甲醇-水(24∶76),检测波长335 nm。结果:从白背叶正丁醇萃取部位分离得到一化合物,鉴定为葫芦巴苷Ⅱ;葫芦巴苷Ⅱ的线性范围为0.053-10.60μg,r=0.9999;平均回收率为99.32%,RSD1.82%(n=6)。结论:该化合物系首次从该植物中分离得到,并首次利用高效液相色谱法建立了测定白背叶中葫芦巴苷Ⅱ含量的方法,方法简便、准确可靠。
Objective: To investigate the chemical constituents and establish a quantitative method of MaUotus apelta. Method: Compound was isolated by silica gel, Sephadex LH-20 column chromatography and Pre-HPLC chromatography. Its structure was identified by physicochemical properties and spectral evidences. The content of M. apelta was determined by HPLC. Chromatographic conditions included Inertsil ODS-3 C18 column (4. 6 mm× 150 mm, 5 μm) and the mobile phase consisting of a mixture of methanol-water (24: 76). The detection wavelength was set at 335 nm. Result: One compound was isolated from n-butanol extract of the M. apelta and its structure was identified as vicenin Ⅱ. The calibration cure was linear in the range of 0. 053-10. 60 μg ( r = 0. 999 9), the average recovery was 99. 32%, RSD 1.82% ( n = 6). Conclusion: The compound was isolated from this plant for the first time. The method to determine the content of vicenin Ⅱ by HPLC was established for the first time. This method is simple, accurate and reliable.
