1型糖尿病大鼠支气管肺泡灌洗液及外周血Th1/Th2细胞因子变化的实验研究

Experimental study of Th1/Th2 imbalance in serum and BALF of the type 1 diabetic mellitus rats

摘要 目的观察1型糖尿病大鼠外周血及支气管肺泡灌洗液(BALF)中Th1/Th2细胞因子的变化。方法20只SD大鼠分为实验组(糖尿病组)和对照组(正常组)各10只,采用链脲菌素制作1型糖尿病大鼠模型,用ELISA法检测血清和BALF中IFN-γ、IL-4水平及IFN-γ/IL-4比值。结果与正常组比较,糖尿病组血清和BALF中的IFN-γ显著升高(P<0.01),IL-4显著下降(P<0.05).IFN-γ/IL-4明显升高(P<0.01)。结论1型糖尿病大鼠血清及BALF中存在着IFN-γ升高,IL-4降低,IFN-γ/IL-4升高,即Th1/Th2失衡。Th1/Th2失衡可能是1型糖尿病易致细菌感染和结核杆菌感染的原因之一。 Objective To investigate the Th1/Th2 imbalance in serum and BALF(bronchoalvedarlavage fluid) of type i diabetic mellitus rats. Methods Twenty SD rat were randomly divided into 2 groups( n = 10 each), the diabetic model group and the contrast group. Type 1 diabetic rat models were induced by strepozotocin. Enzymelinked immunosorbant assay(ELISA) was empolyed to measure interferon-γ(IFN-γ) level,interleukin-4 (IL-4) level in serum and BALF and the ratio o5 IFN-γ/IL-4. Results Compared with normal contrast group,the IFN-γ level o5 the diabetic model group was increased significantly(P 〈 0.01 ), whereas IL-4 level was decreased significantly(P〈 0.05),the ratio o5 IFN-γ/IL-4 was increased(P〈 0;01).Conclusions Thl/Th2 disbalance exists in type 1 diabetes rat Which present as IFN-γ and IFN-γ/IL-4 heighten, IL-4 decreases. Thl/Th2 disbalance perhaps is one o5 the reasons for diabetes mellitus patients bacterial-infection and mycobacterium tuberculosis-infection.
