大孔吸附树脂纯化射干总异黄酮的工艺考察 被引量:4

摘要 目的:筛选分离射干总异黄酮的最佳树脂,并对影响分离的各种因素进行系统的研究,使分离工艺达到最优化。方法:研究7种不同类型吸附树脂对射干总异黄酮的吸附特性,用紫外分光光度法测定射干总异黄酮的含量对工艺进行评价。结果:AB-8型大孔树脂对射干总异黄酮的吸附性能较好,其吸附纯化条件为射干提取物上样浓度为4.0mg.mL-1,吸附流速为2BV.h-1(树脂床体积),以60%乙醇作为洗脱剂进行洗脱效果最佳。经AB-8处理后的射干总异黄酮含量可达60%以上。结论:该法简单可行,分离效果好,能满足于大生产的要求。 OBJECTIVE To screen the optimal separation of the total isoflavones from Rhizoma Belamcandae by selecting appropriate macroporous resins and to study systematically the factors which affect separation. METHODS To study 7 kinds of macroporous resins for isolation and purification of the total isofavones from Rhizoma Belamcandae, and evaluated the separating efficiency by measuring the concentration of total isofavones in Rhizoma Belamcandae with UV spectrophotometer. RESULTS The AB-8 macroporous resin had the best separating efficiency when the concentration of the sample of Rhizoma Belamcandae extract was 4. 0mg/mL, the current velocity of adsorption was 2BV/h (resin bed volume), and 60% ethanol as eluant solvent. The total isoflavones's purity could reach 60%. CONCLUSION This method is simple and feasible with good effect of separation, which can meet industrial requirements.
