冠心康对高同型半胱氨酸血症兔主动脉Bcl-2蛋白表达的影响 被引量:5

摘要 目的探讨高同型半胱氨酸血症(Hhcy)致动脉粥样硬化(AS)时兔主动脉Bcl-2的变化及中药冠心康的防治作用。方法30只健康雄性新西兰大白兔,随机分3组:对照组,模型组,干预组,每组10只,喂食高蛋氨酸饲料方法建立兔Hhcy模型。用循环酶法检测同型半胱氨酸(Hcy)的含量、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量。8周时动物处死,免疫组织化学方法检测兔主动脉Bcl-2蛋白的表达。结果冠心康在降低Hcy的同时,也降低TC、TG、LDL-C的含量(P<0.01),升高了HDL-C,能促进Bcl-2蛋白的表达(P<0.01)。结论冠心康可以抑制内皮细胞的凋亡,对AS的发生发展可起预防和治疗作用。 Objective To explore the change of Bcl-2 in aorta of rabbit model of hyperhomocysteinemia(Hhcy) and the protective effect of herbal medicine Guanxinkang. Methods Thirty rabbits were randomly divided into three groups : the control group, the Hhcy model group, the Guanxinkang intervention group. The model of rabbits with Hhcy was established by feeding high methionine diet. Serum TC, TG, LDL-C, HDL-C and plasma homocysteine were detected with cyclophorase method. Bcl-2 was measured with immunohistochemistry. Results As compared with those in the model group, the expression of Bcl-2 in the Guanxinkang intervention group was markedly increased( P 〈 0.01), the levels of TC, TG, LDL-C and plasma homocysteine were significantly lowered( P 〈 0.01), HDL-C was increased ( P 〈 0.01). Conclusions Guanxinkang can inhibit the development of arteriosclerosis by decreasing the levels of Hcy, TC, TG and LDL-C, increasing the level of HDL-C and promoting the expression of Bcl-2.
