摘要
目的:建立反相高效液相色谱法测定人血浆中盐酸伪麻黄碱浓度的方法。方法:采用Dikma C_(18)色谱柱(250 mm×4.6mm,5μm),流动相:乙腈-水(含0.2%三乙胺)=10:90(磷酸调pH 3.3);检测波长:195nm,流速:0.8ml·min^(-1)。结果:盐酸伪麻黄碱浓度在20~800ng·ml^(-1)范围内线性关系良好,最低定量浓度为20ng·ml^(-1),提取回收率在59.65%~72.20%之间,日内和日间RSD分别为1.34%~4.11%和7.92%~9.98%。结论:本法操作简便、快速、准确、重现好,适用于盐酸伪麻黄碱的药动学研究。
Objective: To establish a RP-HPLC method for determination the concentration of pseudoephedrine in plasma. Method: The separation of pseudoephedrine was performed on Dikma C18 column (250 mm ×4.6 mm, 5 μm). The mobile phase was composed of 0.2 triethanolamine solution- acetonitrile = 90:10 (pH = 3.3 ). The detection wavelength was 195 nm and the flow rate was 0.8 ml. min^ -1. Result: The linear range of pseudoephedrine was 20 - 800 ng· ml^ - 1, The minimal detectable concentration in plasma was 20 ng.ml^ -1 ,the recovery rate was 59.65% -72.20% with RSD at 1.34% -4.11% intra-day and RSD at 7.92% -9.98% interday. Conclusion: This method is simple,fast, accurate and reproducible, which can be used as the pharmacokinetic study for pseudo- ephedrine in human body.
