摘要
目的:通过使用几丁糖作为骨形态蛋白(BMP)载体,与羟基磷灰石构建复合物,探讨其生物相容性和成骨能力,以及作为BMP载体的可行性。方法:制备几丁糖/羟基磷灰石、几丁糖/羟基磷灰石/BMP复合材料。建立兔颅顶骨临界骨缺损模型,以几丁糖/羟基磷灰石、羟基磷灰石、空白为对照,使用几丁糖/羟基磷灰石/BMP复合材料修复顶骨缺损。X线片和组织学切片观察2、4、6、8周后骨缺损修复情况,评价其骨缺损修复能力。结果:几丁糖与羟基磷灰石复合BMP具有早期快速释放BMP的能力及较好的操作性、成形性、生物相容性,对机体微环境影响小,表面矿化效果好,能促进骨缺损修复。结论:几丁糖、多孔羟基磷灰石、BMP复合物,具有良好的骨诱导能力,可作为骨替代材料及BMP的缓释载体,是一种有潜力的骨修复材料。
Objective To investigate the feasibility of chitosan-hydroxylapatite-BMP complex as bone substitute material and a carrier of BMP, Methods Porous chitosan-hydro-xylapatite and chitosan-hydroxylapatite-BMP complex were implanted into calvarial defects. On 2, 4, 6 and 8 weeks after operation bone repairing ability was assessed by radiographic and histological observation. Results Chitosan-hydroxylapatite-BMP complex had an excellent manipulation, shaped ability, biocompatibility, a small influence on body microenvironment, a good mineralization effect on surfaces, The complex had better bone repairing ability than others alone. Conclusion Chitosan-hydroxylapatite-BMP complex may be used as a bone substitute material and a carrier of BMP.
