脓毒症时肝肺微血管内皮细胞炎症基因的表达及意义 被引量:2

摘要 采用盲肠结扎穿孔(CLP)建立小鼠多微生物感染脓毒症模型(CPL组),假手术组(sham组)接受同样的手术操作但不行CLP。分别在致伤后3、12 h分离肝、肺微血管内皮细胞,以RT-PCR方法检测肝肺微血管内皮细胞中多种黏附分子、炎症细胞因子基因的表达。结果显示,与假手术组比较,CLP后3 h肝窦内皮细胞和肺微血管内皮细胞的E-选择素、细胞间黏附分子1(ICAM-1)、血管细胞间黏附分子1(VCAM-1)、肿瘤坏死因子(TNF)α-、IL-1β和IL-6基因的表达水平均明显增高(P<0.01),至12 h时,只有E-选择素基因的表达降至基础水平,其他基因的表达仍明显高于sham组(P<0.01)。认为肝肺微血管内皮细胞是脓毒症时肝肺组织中炎症细胞因子的重要来源之一,也是造成脓毒症时肝肺组织损伤的重要因素。
