不同流式细胞检测系统测定T细胞亚群的一致性评价

Method comparison of two systems of flow cytometry

目的根据美国临床实验室标准化委员会(NCCLS)EP9-A2文件对2种流式细胞检测系统进行比对试验,研究不同系统检测结果的一致性。方法收集40例临床样本,分别使用Beckman Coulter公司XL型流式细胞仪和BD公司CANTO型流式细胞仪同时测定样本的T细胞亚群,分析2组结果的相关性和2种系统检测结果的一致性。结果2种检测系统所得的结果相关性良好,不同系统间的预期偏差在允许偏差范围内,可以接受。结论在严格按操作规程进行校准和质控在控的前提下,使用这2种流式细胞检测系统测定外周血T细胞亚群的结果基本一致。

Objective To estimate the bias between two systems of flow cytometry. Methods T lymphocytes were determined in 40 blood samples by two systems of flow cytometry （ Beckman XL and BD CANTO）. The results of two groups were compared with the procedure as NCCLS-EP9-A2 described. Results The results of the two groups were correlated. The expected bias was in the range of acceptable bias. The results of the two methods were consistent. Conclusions Both methods are advanced flow cytometry methods,the results of two groups are correlated. The two methods can be substitutable, and used to perform the same assay.
