摘要
目的对Beckman-Coulter Z2细胞计数仪(简称Z2)的白细胞(WBC)和红细胞(RBC)检测性能进行系统评价。方法根据国际血液学标准化委员会(ICSH)制定的评价标准对Z2检测WBC和RBC性能包括准确度、精密度、线性、抗干扰、重合计数等进行评价,同时将Sysmex F-820和SE-9500与Z2的结果进行比较。结果Z2检测浓度质控物、WBC和RBC的线性良好(r均>0.999),Z2检测标准颗粒、WBC和RBC的批内、批间和总重复性的变异系数(CV)均<2%,检测WBC和RBC的携带污染率均<1%,在室温或4℃条件下可稳定48 h,高三酰甘油(TG)和高胆红素(Bil)对WBC、RBC的检测基本无干扰(P>0.05)。Z2校正WBC和RBC的重合计数偏差均<1%。Sysmex F-820、SE-9500与Z2检测WBC和RBC结果间的相关性良好(r>0.996),且结果间差异无统计学意义(P>0.05)。结论Z2细胞计数仪检测WBC、RBC具有准确、精密、线性好、抗干扰能力强、重合计数偏差小、与其他厂商的血液分析仪检测结果间具有可比性等特点,可作为WBC和RBC计数的参考计数仪并可用于对其他血液分析仪计数WBC和RBC的校准。
Objective To evaluate the performance of the Beekman-Couher Z2 cell counter for white blood cell (WBC) and red blood cell ( RBC ) counting. Methods The performance of the Z2 cell counter such as accuracy, precision, linearity,interference-resistance and coincidence counting was evaluated according to guidelines published by the International Committee for Standardization in Hematology (ICSH) and compared with that of Sysmex F-820 and SE- 9500. Results The results demonstrated excellent linearity for WBC and RBC ( r 〉 0. 999). The Z2 showed excellent precision,with within-run, between-run and overall coefficient of variance (CV) less than 2%. Carryover was generally acceptable( 〈 1% ). WBC and RBC were stable after blood storage for 48 h at 4 ℃ and room temperature(RT). There was minimal interference for WBC, RBC at high concentrations of bilirubin or triglyeeride( P 〉 0.05 ). The bias of coinci- dence correction in WBC and RBC counting with the Z2 was less than 1%. Correlation between the Z2 and Sysmex F-820 or SE-9500 was excellent for WBC and RBC counting( r 〉 0. 996). Conehmians The Beekman-Couher Z2 cell counter has good features of precision, accuracy, linearity, interference and small deviations of coincidence counting and good comparison with Sysmex F-820 and SE-9500, and it could be used to other hematology analyzer for WBC and RBC counting.
