摘要
目的建立血浆中谷氨酰胺的离子色谱分析方法。方法以氨基酸柱(Amino PAC PA10,2 mm×250 mm)为分离柱,用NaOH溶液和醋酸钠溶液作为流动相,以金为工作电极、pH为参比电极的积分安培离子色谱法分离检测血浆中谷氨酰胺浓度。结果血浆中谷氨酰胺浓度为(680.7±8.6)μmol/L,相对标准偏差为1.02%,加标回收率在96%-102%之间。结论该方法具有灵敏度高、精密度好、分离效果好和样品不需要衍生化处理的优点,适合血浆中谷氨酰胺的浓度分析。
Objective To establish an ion chromatography method for the determination of plasma glutamine. Methods The glutamine in plasma was determined by gradient analysis using sodium hydroxide solution and sodium acetate solution as the mobile phase on an Amino PAC^* PA10(2 mm × 250 mm) column with integrated amperometric detector with Au working electrode and pH reference electrode. Results The level of glutamine in plasma was (680.7 ± 8.6) μmol/L, relative standard deviation was 1.02% and recovery rate ranged from 96% to 102%. Conclusions The method can be used to analyze glutamine in plasma with high sensitivity, good precision and good separative effect.
