摘要
目的:建立高效液相色谱法测定环孢素A全血浓度的方法。方法:采用SpherisorbC8(150mm×4.6mm,5μm)分析柱,流动相为甲醇∶水(75∶25,V/V),检测波长214nm,流速1ml/min,柱温为60℃。结果:环孢素A浓度在50ng/ml~2000ng/ml范围内线性良好(r=0.9998),平均方法回收率为99.7%,日内及日间精密度<6%(n=5)。结论:本方法简便、准确、可靠,能较好的满足临床血药浓度监测的需要。
Objective To establish a HPLC method for the determination of cyclosporin A in whole blood. Methods The Spherisorb C 8 column( 150 mm×4.6 mm,5 μm)was used,the mobile phase was methanol-water(75:25, V/V) ,the detection wavelength was at 214 nm, the flow rate was 1 ml/min and column temperature was 60 ℃. Results The linear range of cyclosporin A was 50 ng/ml ~ 2 000 ng/ml( r = 0.999 8 ). The average recovery was 99.7% ( n = 5 ). Within-clay and betweenclay variation co-efficients were less than 6%. Conclusion The method is simple, accurate, and reliable. It is suitable for therapeutic drug monitoring.
