醒髓汤对实验性大鼠脊髓损伤组织中降钙素基因相关肽的影响 被引量:5

Effect of Xingsui decoction(醒髓汤)on calcitionin gene related peptide(CGRP)of spinal cord injury in rats

摘要 目的:研究醒髓汤对大鼠急性脊髓损伤治疗作用及对神经组织中降钙素基因相关肽(CGRP)的影响。方法:48只大鼠随机分为3组,空白对照组20只,模型组8只,醒髓汤组20只。用改良Allen′s法造脊髓损伤模型。空白对照组仅咬除棘突,剥离椎板,正常饲养;模型组造模后灌生理盐水;醒髓汤组灌中药醒髓汤。于手术后2、6、24 h用斜板试验评价大鼠的神经功能,并取脊髓匀浆,测量脊髓中CGRP的含量。结果:模型组CGRP在造模成功后逐渐上升,6 h达到顶峰,24 h恢复至正常水平。醒髓汤组与模型组伤后2 h无明显差别,CGRP均升高,6 h后达到顶峰,两组与空白对照组相比有显著性差异(P<0.01)。而醒髓汤组在6 h后匀浆组织中CGRP含量高于模型组(P<0.05)。醒髓汤组与模型组于伤后6 h在神经功能恢复方面无显著性差异。但在24 h后醒髓汤组与模型组Travor评分有显著性差异(P<0.05)。结论:醒髓汤对大鼠脊髓损伤有较好的治疗作用,并可增加脊髓中CGRP的含量。 Objective:To study the therapeutic effect of Xingsui decoction on spinal cord injury( SCI )and the effect on the level of CGRP of spinal cord in rats. Methods:Forty-eight SD rats were randomly divided into 3 groups ,20 rats in the blank group,8 in the model group and 20 in Xingsui decoction group. The rats model was made by Allen's weight drop method. Rats in blank control group were only cut off spinal process and decoherenced vertebral plate, and were fed normally. After models were made successfully,rats in model group were fed with saline and rats in Xingsui decoction group were fed with Xingsui decoction. The nerve function was messured on the inclined plane test and the content of CGRP in spinal cord was determined at 2,6,24 hours after injury by the Allen's method. Results:In model group, the content of CGRP in spinal cord increased significantly after injury and reached the zenith at 6 hours and recovered the normal level at 24 hours. Xingsui decoction group had no significant difference as compared with model group at 2 hours and CGRP increased and reached the zenith at 6 hours. There were significant difference on CGRP between these two groups and the blank control group. The content of CGRP in Xingsui decoction group at 6 hours after injury was higher than that of model group. There were no significant difference on recovery of nerve function between Xingsui decoction group and model group at 6 hours after injury. But at 24 hours after injury there were significant difference on Travor scoring between these two groups. Conclusion :Xingsui decoction has good therapeutic effect on spinal cord injury,and increase the content of CGRP in spinal cord.
