摘要
目的观察侧脑室注射链脲佐菌素对大鼠脑PSD-95和Shank1表达的影响。方法将大鼠随机分为正常对照组和模型组,模型组大鼠双侧侧脑室注射链脲佐菌素3mg/kg,第3d重复此剂量;对照组以人工脑脊液代替链脲佐菌素。21d后,取大鼠海马,用免疫组织化学和Western blotting方法观察突触相关蛋白PSD-95和Shank1的表达。结果与对照组相比,模型组大鼠海马PSD-95和Shank1阳性细胞明显减少,PSD95和Shank1蛋白表达降低。结论侧脑室注射链脲佐菌素使海马PSD-95和Shank1表达减少,干扰了神经元突触信号传导。
Objective To investigate the effects of intracerebroventricular(ICV) administration of streptozotocin (STZ) on expression of PSD-95 and Shankl in rat hippocampus. Methods A total of 30 rats were randomly divided into normal control group and model group. The rats in the model group received intracerebroventricular injection of STZ bilaterally with a dose of 3mg/kg and the injection was repeated on the third day. The rats in normal control group received the same treatment as the first group except for STZ injection. After 21 days, the brains of the rats were taken, frozen sections of brain were prepared, and then PSD-95 and Shankl were stained with immunohistochemistry. Furthermore, the contents of PSD-95 and Shankl were detected by Western blotting. Results After ICV injection of STZ, the expression of PSD-95 and Shankl positive cells and the contents of PSD-95 and Shankl in hippocampus both decreased. Conclusion ICV injection of STZ can lead to the decrease of the expression of PSD-95 and Shankl, indicating it may damage the neuron's synaptic signal transduction.
