摘要 以[Mo17(rf_3rf_3)CMS-唐徐×HZ_1(Rf_3Rf_3)N]×Mo17(rf_3rf_3)N回交群体作为基因定位群体。采用RFLP和RAPD分子标记技术,定位了Rf_3/rf_3基因。RFLP分析表明,Rf_3/rf_3基因位于第二染色体长臂上的UMC36A和UMC49分子标记之间,其遗传距离分别为12.7cM和4.8cM。采用BSA法,筛选了340个10mer随机引物,找到与Rf_3基因紧密连锁的分子标记RAPD Eo8-1.2,将Rf_3/rf_3基因的标记距离缩小到2.7cM,为Rf_3/rf_3基因辅助选择提供了有价值的分子标记。 To locate the Rf3 gene that can restore the pollen fertility of CMS-S maize, a backcross [Mol7(rf3rf3)CMS-Tangxu×HZ1(Rf3Rf3)N]×Mol7(rf3rf3)N]was used as mapping population. RFLP analysis shown that the Rf3/rf3 locus is flanked by the markers UMC36A and UMC49 on the long arm of chromosome 2, with map distances of 12. 7 cM and 4.8 cM, respectively. In RAPD analysis, 340 10-mer random primers were screened through sterile and fertile bulked DNAs, and one RAPD marker produced by OPE08 was found to be closely linked to Rf3/rf3 gene. RFLP and RAPD data were analyzed by the program of Join-MAP. The result indicated that RAPDE08-1. 2 marker was located between UMC36A and Rf3/ rf3, with a map distance of 2. 7 cM from Rf3/rf3 locus.
