双载体法固定化黑曲霉发酵产壳聚糖酶的研究 被引量:8

摘要 探索玉米芯颗粒吸附、海藻酸钠包埋固定化黑曲霉(Aspergillus niger)孢子的方法以及双载体固定化黑曲霉发酵产壳聚糖酶的条件.实验结果表明,玉米芯颗粒对黑曲霉孢子的吸附容量约为5×107个/克;适宜包埋剂质量分数为1·0%;0·2g玉米芯颗粒经吸附、包埋与增殖预培养成熟的固定化黑曲霉细胞发酵产壳聚糖酶最适pH为5·0,适宜发酵装液量为25mL(250mL三角瓶),28·6℃,132r/min条件下摇床发酵48h产壳聚糖酶活力101·3U/mL. Mycelia of Aspergillus niger were absorbed with corncob granule and embedded with sodium alginate. The Immobilization of Aspergillus niger cells was applied in chitosanase production. The max absorbability of corncob granule to the spores of A.niger was 5 X 107/g. The optimized w(sodium alginate) was 1.0%;The optimized pH for chitosanase production by immobilized A. niger cells absorbed with 0.2 g corncob granules and embedded with sodium alginate was 5.0. After 48 hours'fermentation at 132 r/min and 28. 6℃,the chitosanase activity produced by the mycelia of A. niger was as high as 101.3 U/mL.
