摘要
利用300MPa的高静水压处理黏红酵母(Rhodotorula glutinis)RY06菌株10min,获得一株突变株RY06-27,其β-胡萝卜素产量从原来的6.30mg/L增加到9.64mg/L,提高了53%。通过响应面分析法优化发酵培养基,可使该突变株β-胡萝卜素产量达13.43mg/L,其优化培养基组成为酵母膏4.23g/L、接种量30mL/L、番茄汁2.5mL/L、培养基装量40mL/250mL、花生油0.5mL/L、pH6.7、葡萄糖12.11g/L、硫酸铵5g/L。
The strain Rhodotorula glutinis RY06(β-carotene of 6.30 mg/L) was subjected to mutagenesis for strain improvement by high hydrostatic pressure(HHP), and mutant strain RY06-27 was selected out. The yellow colored mutant RY06-27 (obtained at 300MPa for 10min) produced 1.53 fold more β-carotene (9.64mg/L) than the parent strain RY06. The response surface methodology(RSM) for medium optimization for β-carotene production by strain RY06-27 was studied by a factorial design and a central composite design. The maximum yield of β-carotene (13.43mg/L) is obtained with the optimum medium at yeast extract 4.23g/L, glucose 12.11g/L, pH6.7, volume of inoculums 30mL/L, tomato exact 2.5mL/L, volume of media 40mL/250mL, peanut oil 0.5mL/L and (NH4)2SO4 5mL/L.
