游离移植后长期存活的兔颌下腺细胞的生物学特性

摘要 目的探讨游离移植后长期存活的兔颌下腺细胞的生物学特性。设计实验性研究。研究对象兔颌下腺细胞。方法将移植成功后存活8个月的兔取出移植的颌下腺,进行细胞培养,观察其细胞生长特性,进行流式细胞仪分析及甲基噻唑基四唑(MTT)检测,并与培养的正常颌下腺细胞进行对比。主要指标细胞生长特性、流式细胞仪分析、细胞生长曲线。结果移植颌下腺细胞组与正常颌下腺细胞组具有相似的细胞形态和生长特性,移植颌下腺细胞组细胞周期的G1期、S期及G2期各时相细胞增值百分比分别是(82.7±2.10)%、(6.2±0.98)%、(11.2±1.24)%,正常颌下腺细胞组各时相分别是(82.0±2.06)%、(6.3±1.12)%、(11.7±1.35)%,两组相比较无明显差异(P均>0.05),同时两组具有相似的细胞生长曲线。结论游离移植后长期成活的兔颌下腺细胞与非移植的正常颌下腺细胞具有相近的细胞生长特性,去除神经支配后游离移植的颌下腺细胞的生物学特性未发生明显改变。 Objective To investgate the cytobiological characteristic of rabbit transplanted autologous free submandibular gland (SMG) for long-term surviving. Design Experimental study. Participants Rabbit submandibular gland cells. Methods The cultured cells of rabbit transplanted SMG long-term surviving for 8 months were observed through inverted biological microscopy, flow-cytometry and MTT, then contrasted with the cultured normal SMG cells. Main Outcome Measures The cultured cell growth characteridtic and the cell growth curves. Results The cell percentage of G1 phase, S phase and G2 phase was (82.7±2.10)%, (6.2±0.98)%, (11.2±1.24)% in the transfer SMG cells group, otherwise, was (82.0±2.06)%, (6.3±1.12)%, (11.7±1.35)% in the normal SMG cells group. There was no significant difference between the two groups (P〉0.05). Moreover, the growth curves of the two kinds of cells were similar. Conclusions The cell growth properties of cultured transplanted rabbit SMG are similar to that of normal SMG, the cytobiological characteristic of transplanted autologous free rabbit SMG are not changed evidently.
